Specific network folder takes ~15 seconds to load for all Windows users

At our office, we are having problems with a specific folder on our network.

One particular folder, and no others, takes 15 second or so to load (along with the green progress bar in File Explorer) for every single user, each time they access it, even when going back from its own subfolders. This is universal across all versions of Windows (7, 8.1, and 10) for all users*.

*Our one employee with a Mac does not have this issue, the folder opens immediately for her.

The folder also opens immediately when directly on the server.

The server is running 2008 R2 Standard; it has a file server role, but we recently made it into a domain controller as well.

We have tried the "Optimize the folder for General Items" trick but this does not work.

We access this folder frequently, and the slow-down interrupts the workflow of our employees
